Optimization of Culture Conditions for Keratinase Production in Streptomyces Sp. JRS19 for Chick Feather Wastes Degradation
نویسندگان
چکیده مقاله:
The aim of this study was to find the optimal conditions of a newly isolated Streptomyces strain JRS 19. Screening for keratinase producing bacteria and their keratinase production were investigated. The keratinase produced by the isolate was purified by chromatographic techniques and the optimum pH maintained was 7.0 and maintained at different temperatures Viz., 20ºC, 25ºC, 30ºC, 35ºC and 40ºC. Besides temperature and pH ,the enzyme is active upon different sources and its effect on carbon, nitrogen , aminoacids were determined and recorded the protein content and enzyme activity. The carbon sources revealed that sucrose was found highly significant, whereas peptone was found highly significant in nitrogen source. The effect of keratinase on amino acid shows that valine was found highly significant. Based on this study, the physio-chemical properties of keratinase were analyzed and the results were interpreted. [T. Jayalakshmi et al. results the pcr approved the bacillus genus of the isolates. the strain of bacillus subtilis was identified using biochemical tests. 40 ºc and ph 11 are the optimum condition for maximum keratinase enzyme activity. objectives the aim of this study was the isolation of feather degrading bacillus spp.from a poultry waste and the optimization of conditions for the highest enzyme activity and feath...
متن کاملEfficient Degradation of Feather by Keratinase Producing Bacillus sp.
Keratinase producing microorganisms are being increasingly utilized for degradation and recycling of poultry feather waste. Two native strains BF11 (Bacillus subtilis) and BF21 (Bacillus cereus) degrading keratin completely were characterized. The native strains produced more than 10 KU/mL of enzyme. Strain improvement resulted in isolation of MBF11 and MBF21 from BF11 and BF21 isolates, respec...
متن کاملScreening of fungi efficient in feather degradation and keratinase production
Two hundred thirty four fungal strains were isolated by baiting method, for their feather degradation and keratinase producing ability. Fungi were tested on solid milk agar plates and in submerged culture. Maximum clearing zone was made by Chrysosporium indicum (7mm) on solid agar plates. The highest keratinase production was found in case of Acremonium strictum (74.40Unit/mL & 124.72 Unit/ml i...
متن کاملOptimization of Culture Conditions and Medium Components for Carboxymethyl Cellulase (CMCase) Production by a Novel Basidiomycete Strain Peniophora sp. NDVN01
Background: Cellulytic enzymes including carboxymethyl cellulases play the key role in hydrolysis of cellulose, a huge organic carbon reservoir on earth, into monomeric sugars and their eventual conversion into valuable chemicals and energy sources. Objectives: In this study, we described the identification of a basidiomycete isolate NDVN01 and optimization of culture conditions and medium com...
متن کاملOptimization of culture conditions for growth of the Aurantiochytrium sp. shy, isolated from the Persian Gulf
In this study, a native microalgae strain with a high capability of DHA production has been isolated and purified from mangrove shores in the Persian Gulf. Morphological and molecular identification based on 18S rRNA gene showed that the isolated strain belongs to Aurantiochytrium genus. In order to optimize the growth of this strain, various environmental factors were examined, including tempe...
